Effects of the surface charge on the stability of PEG-b-PCL micelles: simulation of the interactions between charged micelles and plasma components

We reported the effects of the apparent surface charge on the enzymatic degradation behavior of PEG-b-PCL micelles. We simulated the interactions between charged micelles and plasma components and investigate the stability of surface charged micelles in the presence of BSA. We synthesized three types PEG-b-PCL di-blocks containing CH3O, NH2, and COOH groups, respectively, at the end of the PEG block. These copolymers can self-assemble into uniform micelles bearing different surface charges. The surface charge of micelles can be simply adjusted by changing the feed ratio of NH2-PEG-b-PCL and COOH-PEG-b-PCL. Mixed charged micelles showed good stability in the presence of BSA and enhanced cell uptake was also expected given the zwitterionic characteristic of the micelle surface. With the tunable surface charge of micelles, we can design novel drug delivery carriers which possess both long blood circulation and efficient cell uptake.
